We are seeking a highly motivated cell biologist or immunologist to join the Immuno-oncology Discovery team at Merck in Boston to help us bring the next generation of cancer therapeutics to patients. You will be responsible for executing and reporting the results of experiments to support projects from target identification and validation through to clinical candidate selection, as well as supporting the development and clinical pipelines with basic biology and pharmacology studies.

You will use a wide range of advanced methodologies to probe mechanisms and test drugs, primarily through in vitro and ex vivo cell-based assays. The successful candidate will have excellent oral and written communication skills, work well independently and as part of a team. The role includes developing and optimizing methodologies and assays and making decisions using good scientific judgment. The position is a hands-on, laboratory-based role with no direct reports.

Qualifications

Education:

- BS/MS degree in immunology, cell biology, pharmacology, or related field.

Required:

- Cell line and primary immune cell tissue culture skills.

- Ability to design and perform experiments and interpret data derived from cellular immune function assays (e.g. differentiation, proliferation, cytotoxicity, cytokine production).

- Familiarity with assays and technologies commonly used to assess immune endpoints (e.g., MLR, antigen-specific T/APC cultures, ELISA, Mesoscale).

- Familiarity with assays and technologies commonly used in cell biology and cell signaling studies, e.g. Western blot, immunoprecipitation, Ca flux imaging, receptor-ligand pharmacology.

- Ability to organize and communicate complex data sets in a clear and concise manner.

- Basic understanding of data analysis, and assessment of reproducibility and reliability of experimental data.

Preferred:

- Cell line engineering for assay development, molecular biology, PCR technologies, genetic manipulation.

- Experience in cellular assay development, tissue culture, primary human and in vivo cell culture, RT-PCR or Nanostring, flow cytometry heavily preferred.

- Directing and coordinating work contract research organizations for routine work such as cloning or vector construction.

- Proven experience with biologics drug discovery and demonstrable knowledge of multiple large molecule formats and associated technologies

- Demonstrable experience with handling and in vitro pharmacological characterization of small molecule drugs, e.g. potency titration, experimental design and replication.

- Experience authoring peer-reviewed scientific publications and / or presenting at scientific conferences, particularly in the field of immuno-oncology.

- Isolation, characterization and functional testing of primary human immune cells from healthy donors or patient-derived materials.

- Experience with cell separation technologies (e.g., Miltenyi, AutoMACS) and FACS sorting.

- Experience in multi-parameter flow cytometry and proficiency in downstream FACS data analysis programs such as FlowJo, FACSDiva, etc.

- Experience with CRISPR, either on a single gene basis or screening in phenotypic assays.

- Experience with RNA profiling technologies – RT-QPCR, Nanostring, RNAseq and in particular single cell profiling.
