We are looking to appoint a Research Associate in the School of Physiology, Pharmacology & Neuroscience to work on a molecular mediator of platelets’ procoagulant membrane dynamics and thrombosis.

Platelets are critical mediators of the haemostatic response. We have recently shown that they undergo a remarkable transformation in shape, upon interaction with collagen, which we call ballooning and procoagulant-spreading. We showed this transformation to be important in amplifying the procoagulant response of platelets, by markedly increasing their surface area, and thereby important for thrombus formation in vivo. The ballooning and procoagulant-spreading is dependent upon fluid entry into the cells, and we hypothesize that the water channel aquaporin-1 (AQP1) is the critical water entry system.

The main purpose of this job is to elucidate the role of AQP1 in haemostasis and platelet driven thrombosis. We propose a series of experiments to address how aquaporin-1 is regulated in platelets upon activation, how it may control the function of platelets and how it may regulate the procoagulant response and thrombus formation, in both the arterial and venous circulations. Understanding the role of AQP1 in platelets may provide a very novel approach to controlling ischemic vascular disease.

The project will be significantly imaging based but will also involve the use of multiple biochemical, proteomic and cell biology approaches in platelets, using mouse gene knockout models. You will use state-of-the art imaging facilities, light and electron microscopy, to examine subcellular localization, kinetics and mechanism of membrane dynamics and AQP1 regulation of these events. Accordingly, practical experience in biochemical, and platelet imaging techniques is essential and you should have a strong and proven interest in cell biology.

In addition you should have a degree in Biochemistry, Cell Biology or related subject and a PhD in Biochemistry/Cell or Molecular biology or related subject.
