This project aims to accurately measure immature platelets for the first time in health and disease. Immature platelets or “preplatelets” will be measured using Imagestream flow cytometry in parallel with conventional flow cytometry, the Immature Platelet Fraction (IPF) and confocal microscopy (on blood films/slides). Comparisons will be made in EDTA and citrate anticoagulated whole blood/PRP labelled with anti-CD61, anti-tubulin and nucleic acid dyes. The percentage/absolute number of the immature preplatelets will be measured in samples from normals and inherited or acquired forms of thrombocytopenia where platelet production is abnormal. We anticipate that this study will result in the development of a new method of accurate immature preplatelet enumeration with obvious potential for clinical translation as well as increasing our understanding of the changes of platelet kinetics in disease.

Person Specification
Applicants should have a strong background in Biology, and ideally a background/interest in Flow Cytometry, microscopy and Image analysis. They should have a commitment to research in Haematology/ Platelet Biology and hold or realistically expect to obtain at least an Upper Second Class Honours Degree in a relevant subject.
