Thesis description

In most species, meiosis requires a specific program for the formation and repair of double-stranded DNA (DSB) breaks by homologous recombination called meiotic recombination. It is a fundamental part of sexual reproduction that plays a key role in the segregation of chromosomes during the first meiotic division. Faults in meiotic recombination can cause segregation errors that can lead to various pathologies, such as infertility or aneuploidy.

It is known that the formation of DNA double-strand breaks requires the Spo11 catalytic complex as well as several meiotic factors suggested to form a complex called recombinosome, but the molecular details underlying this important process are not known. The goal of this doctoral project is to elucidate the molecular architecture of meiotic recombinosome and analyze its interaction with the Spo11 catalytic complex to understand how these factors work together to generate and regulate DNA double-strand breaks. . The structure, function and interactions of these factors will be studied in an interdisciplinary approach combining biochemistry, biophysics and structural biology (X-ray crystallography, cryo-EM) with the in vivo characterization of mouse models to provide new insights into this important mechanism responsible for increasing genome diversity. The project will be carried out in collaboration with a group of cell biology / genetics at the Montpellier IGH.

Work context

The Institute of Structural Biology (IBS) aims to understand basic biological mechanisms and human diseases at the molecular and structural levels. Its themes are developed in 16 groups, bringing together about 270 people including 30 doctoral students, who pursue their research in a multidisciplinary way, at the frontiers of biology, physics and chemistry. 

Located within the European Photon and Neutron (EPN) Campus in Grenoble, the IBS is part of a larger ensemble, the Partnership for Structural Biology (PSB), which offers state-of-the-art facilities for innovative research in the sciences. biomolecular. EPN is part of the GIANT Science Park, a network of six centers of excellence with more than 6,000 researchers.

The IBS is attached to the Graduate School of Chemistry and Life Sciences at the University of Grenoble Alpes (formerly known as the Joseph Fourier University), which ranks among the top 100 of the international ranking of Shanghai universities in the field of natural sciences.

Constraints and risks
